If you are one of the many who use iTunes for a library AND a store, these tips will really be enjoyable for you.
1. Get it while it's free. - Once every week, the iTunes store will release a "new" song for digital download for free. These songs are often not HUGE name artists, but on several occasions, one song has been all it takes for an artist to get popular. I suggest listening to all the free songs, and if you find one you like, get it while it's free; lest you have to pay the $1.29 that popular songs cost.
2. DJ McEinstein - One of the greatest features of iTunes lately has been the "Genius" feature. This feature anonymously sends data about your music library to Apple's servers where it is then analyzed and "teaches" iTunes about your favorites, correlations between genres and artists, enabling iTunes to create GODLY playlists and to find music that you ACTUALLY will like for purchase via the sidebar.
3. iTouched iT. - More often than not, if a very useful free app is released and becomes popular, the developer will try to capitalize on it by adding a HUGE amount of features, and adding a pricetag to it, in the hopes that future users will pay and get the upgrade. Look for free "do it all" or "multipurpose" apps and download them. If you don't want to crowd your precious springboard real estate, you can delete them from the library, but iTunes will remember that you've bought this before and will allow you to download the update for free.
Just these few tips should save you some cash, and allow you to have a more enjoyable listening experience too.
